چکیده

Glutathione (GSH), a tripeptide (L γ-glutamyl-cysteinyl-glycine) is the major nonprotein thiol found abundantly in eukaryotic cells (1–3). GSH plays a critical role in many metabolic pathways. It acts as a cofactor for intracellular enzymes like the glutathione S-transferases (GST) and glutathione peroxidase; it aids in amino acid transport and is essential for detoxification of xenobiotics and reactive oxygen species (ROS). GSH is also involved in maintenance of cellular redox-regulated signal transduction. Reactive oxygen species and unintended drugdrug interactions often cause a drop in GSH levels by reaction DETECTING TOXICOLOGICAL RESPONSES IN CELLS WITH THE BIOLUMINESCENT GSH-GLOTM GLUTATHIONE ASSAY

The role of the glyoxalase pathway in reducing mesothelial toxicity of glucose degradation products.

BACKGROUND The glucose degradation products (GDP) presentin conventional peritoneal dialysis fluids (PDF) may exert adverse effects toward human peritoneal mesothelial cells (HPMC). Some GDP can be detoxified by the glyoxalase/ glutathione pathway. It has been shown that the addition of glyoxalase I (GLO-I) and reduced glutathione (GSH) to PDF effectively eliminates GDP. We have therefore exami...

Reduced glutathione alleviates tunicamycin-induced endoplasmic reticulum stress in mouse preimplantation embryos

Endoplasmic reticulum (ER) stress, a dysfunction in protein-folding capacity, is involved in many pathological and physiological responses, including embryonic development. This study aims to determine the developmental competence, apoptosis, and stress-induced gene expression in mouse preimplantation embryos grown in an in vitro culture medium supplemented with different concentrations of the ...

Intracellular GSH Alterations and Its Relationship to Level of Resistance following Exposure to Cisplatin in Cancer Cells

One of the major complications in cancer chemotherapy is the development of resistance, and cisplatin, as one of the important medicines in treatment regimens of different cancers is not excluded. One of the most described cellular defense mechanisms involved in resistance is glutathione (GSH) and in this study, the effects of cisplatin on the total intracellular GSH level (GSHi) in some sensit...

Genotoxicity of 4-hydroxy-2-nonenal in human colon tumor cells is associated with cellular levels of glutathione and the modulation of glutathione S-transferase A4 expression by butyrate.

The cellular production of 4-hydroxy-2-nonenal (HNE), a product of endogenous lipid peroxidation, constitutes a genotoxic risk factor for carcinogenesis. Our previous studies have shown that human HT29 colon cells developed resistance toward HNE injury after treatment with butyrate, a diet-associated gut fermentation product. This resistance was attributed to the induction of certain glutathion...
